Lion's Mane + Psilocybin Interaction
⚠️ RESEARCH-BASED INTERACTION DATA — NOT COMPREHENSIVE.
Interaction Type: synergistic | Evidence Level: preliminary
The 'Stamets Stack' — lion's mane + psilocybin microdose + niacin — combines NGF stimulation with 5-HT2A-mediated neuroplasticity for enhanced cognitive and neurorestorative effects.
Mechanism of Interaction
Lion's mane contains hericenones and erinacines that stimulate nerve growth factor (NGF) synthesis and secretion. Psilocybin (via psilocin) activates 5-HT2A receptors, triggering BDNF release and dendritic spine growth. NGF and BDNF support complementary aspects of neuroplasticity — NGF for nerve regrowth, BDNF for synaptic strengthening. Niacin (in the Stamets protocol) promotes peripheral vasodilation, theoretically enhancing CNS delivery.
Clinical Relevance
Based on Paul Stamets' patent and anecdotal reports. No published clinical trials of this specific combination. Lion's mane alone has small RCT data showing cognitive improvement. Psilocybin has Phase II/III data for depression. The combination is widely discussed in neurohacking communities.
Recommendations
- Lion's mane: 500-1000mg daily (hot water or dual extract)
- Psilocybin microdose: sub-perceptual (0.1-0.3g dried mushroom equivalent)
- Niacin (flush form): 100-200mg
- Legal status of psilocybin varies — research use only in most jurisdictions
