Hesperidin for Alzheimer's Disease

Hesperidin crosses the BBB and provides neuroprotection through anti-inflammatory and antioxidant mechanisms.
Mechanism of Action
Hesperidin inhibits acetylcholinesterase (similar to donepezil mechanism), activates TrkB/BDNF signaling, reduces amyloid-beta aggregation, and suppresses NF-κB-driven neuroinflammation. It also improves cerebral blood flow.
General mechanism: Citrus flavanone glycoside. AChE inhibitor, BDNF/TrkB activator, NF-κB inhibitor, Nrf2 activator. Crosses BBB.
Current Evidence
Preclinical AD models show improved cognition and reduced pathology. Epidemiological data links citrus flavonoid intake to reduced dementia risk. Clinical trials for cognitive health ongoing.
Clinical Status: Preclinical for AD. Phase I/II for cognitive health.
