Honokiol for Stage IV Cancer
Also known as: Magnolia bark extract
Honokiol is a potent anti-cancer compound from magnolia bark that crosses the BBB, making it unique for brain tumors.
Mechanism of Action
Honokiol activates mitochondrial apoptosis, inhibits mTOR/STAT3/EGFR signaling, suppresses NF-κB, and blocks epithelial-mesenchymal transition (anti-metastatic). Its BBB penetrance makes it valuable for glioblastoma and brain metastases.
General mechanism: Biphenol from magnolia bark. SIRT3 activator, NF-κB/STAT3 inhibitor, mTOR suppressor. Crosses BBB/BSCB.
Current Evidence
Preclinical evidence strong for glioblastoma, breast, prostate, and lung cancer. BBB penetrance confirmed. Synergy with temozolomide for GBM demonstrated. Phase I studies planned.
Clinical Status: Preclinical. Phase I planned for glioblastoma. Available as supplement.
Safety Profile
Safe in preclinical studies. Long history of use in traditional medicine. Mild sedative effects. Limited human safety data at therapeutic doses.
